We have over 450 trees on 5/16 tubing. Without changing all the 5/16 laterals. Would it be beneficial to change the drops to 3/16? Would that give us more vacuum at the tap?

Replacing your drops will give you more sap because of no bacteria but this won't create any more vacuum.

Well, it might give you 1-2" of vacuum, not worth it. If you have good slope, especially as the lateral approaches the main line or a tank, you would be best off, removing the 5/16 and putting in the 3/16. I did just that on shy of 200 taps and their performance is incredible. Far better than my taps that are on 5/16, and both feed into mains with a vacuum pump on them at 19" vac. On my 3/16 laterals I have 25-35 taps/line.
